Peptides are messengers, and the marketing runs ahead of the evidence

Short chains of amino acids that signal within skin are a genuinely interesting idea. The gap between that idea and a finished cream is where the claims get loose.

What holds up in practice

What a peptide is

A peptide is a short chain of amino acids, shorter than a protein, and skin uses many of them as signals between cells. Cosmetic peptides are designed to mimic fragments of larger molecules that appear during processes such as wound repair or collagen turnover. The reasoning is that presenting a fragment associated with repair might prompt cells to behave as though repair were needed.

That is an elegant idea and it is why the category has attracted so much formulation effort over the past two decades. Whether it works reliably on intact skin through a cream is a separate and much harder question.

The penetration problem

The outer layer is specifically built to keep water-soluble molecules out, and peptides are water soluble and comparatively large. Formulators attach fatty chains, use delivery systems or shorten sequences to improve the odds, with varying and rarely published success.

After four to six weeks, a peptide that cannot reach living cells cannot signal to them, however elegant its design. This is the reason serious discussion of peptides always circles back to delivery rather than to the sequence itself. Claims that skip the delivery question entirely should be read with that omission in mind.

The four broad types

Signal peptides aim to prompt production of structural proteins, and matrixyl-type sequences are the best known example. Carrier peptides deliver trace elements such as copper into skin, where those elements participate in enzymatic processes. Enzyme-inhibiting peptides interfere with the breakdown of existing structural proteins rather than encouraging new ones.

Neurotransmitter-inhibiting peptides are marketed on an analogy with injectable muscle relaxants, an analogy that is considerably weaker than the packaging implies. That last group carries the strongest claims and, generally, the thinnest independent support.

How strong is the evidence

A good deal of peptide research is conducted in cell culture or on excised skin, which shows a mechanism is possible rather than that a cream works. Clinical work exists but is frequently small, short and funded by the ingredient supplier, which is common in cosmetics and worth knowing. This does not make peptides useless; it makes the confidence level lower than for retinoids or sunscreen.

Being honest about that gradation is more useful than either dismissing the category or repeating supplier claims.

Peptides sit in the plausible-and-under-evidenced tier, alongside a great many fashionable actives.

What to expect in practice

Peptide serums are almost always comfortable, rarely irritating and pleasant to use, which is a real advantage for reactive skin. Any visible improvement is likely to be gradual and modest, and much of the immediate effect is hydration from the base formula. They combine easily with other ingredients, with the occasional caution around very low pH formulas that may affect stability.

Judging them requires months rather than weeks, and honest self-assessment is difficult without photographs in consistent light. If budget is limited, sunscreen and a retinoid have a stronger evidence base for the same concerns.

Patch test anything new, and introduce one active at a time so you can tell what did what.

Reading peptide marketing

Trademarked complex names usually correspond to one or two INCI-listed peptides plus a solvent, and the list will show you which. Language about instructing, reprogramming or reactivating skin is doing rhetorical work rather than describing a measured outcome. Comparisons to injectable procedures are the clearest signal that the copy has left the evidence behind.

Concentrations are almost never disclosed, and peptide position on an ingredient list is usually below the one per cent line. None of this means avoid them, only that expectations should be calibrated to a young and commercially entangled literature.
